An Easter egg hunt is all about spotting what stands out. The brighter the clue, the easier it is to find. Grid colors work the same way. By adding color based on sequence steps, response values, or technical outliers, you can quickly identify patterns and uncover the details that matter.

This time, the eggs are already sorted for you. The plate layout is grouped into four "baskets," each representing a different assay element. Follow the colors to see how each element fits into the assay.

The layout comes from the bioMérieux Endotoxin Detection Assay Package add-on, using a GOPLATE™ layout prepared with the ENDOZYME® II GO assay kit to test the purity of pharmaceutical water.

Standard

The reference standard of the assay document. You define the dilution sequence of the standard as a geometric sequence employing parameters for the number of steps, the initial working concentration in EU/ml, and the base of the sequence.

Blank

The blank control sample that has neither an active ingredient nor an endotoxin added to it. The assay kits from bioMérieux use this sample to adjust the standard in the case of a linear fit. When using a linear model for assay analysis, using a Blank control element is therefore mandatory.

Positive Product Control

The PPC control sample typically only contains dilutant and endotoxin at a known concentration. The endotoxin level measured in this sample is therefore mainly due to the PPC control spike, that is, the amount of endotoxin added to the PPC control. The PPC control sample does not affect the calculation or the validity of the document.

Sample

The sample lets you combine information on the test sample such as dilution factor, endotoxin limits, and number of replicates with its associated PPC. The PPC spike setting indicates the amount of endotoxin added to the otherwise exact same test sample substance.
